Annexin V 是檢測(cè)早期細(xì)胞凋亡的最靈敏指標(biāo)之一。正常細(xì)胞中,磷脂酰絲氨酸 (PS)只分布在細(xì)胞膜磷脂雙分子層的內(nèi)側(cè),而在細(xì)胞凋亡早期,細(xì)胞質(zhì)膜內(nèi)側(cè)的磷脂酰絲氨酸發(fā)生外翻,暴露于細(xì)胞質(zhì)膜的外側(cè)。Annexin V是一種與磷脂酰絲氨酸有高度親和力的Ca2+依賴性磷脂結(jié)合蛋白,因此能夠利用Annexin V與細(xì)胞外側(cè)暴露的磷脂酰絲氨酸相結(jié)合的性質(zhì),檢測(cè)細(xì)胞的早期凋亡。將 Annexin V進(jìn)行綠色熒光 (FITC) 標(biāo)記,以標(biāo)記了的Annexin V-FITC 作為探針,利用流式細(xì)胞儀或熒光顯微鏡可檢測(cè)細(xì)胞凋亡的發(fā)生。碘化丙啶 (PI)是一種核酸染料,它不能透過(guò)具備完整細(xì)胞膜的正常細(xì)胞和早期凋亡細(xì)胞,但能夠透過(guò)凋亡中晚期的細(xì)胞和壞死細(xì)胞的細(xì)胞膜而將細(xì)胞核染色,在特定激發(fā)光條件下呈現(xiàn)紅色。
將 Annexin V-FITC與 PI 匹配使用,就可以將處于不同凋亡時(shí)期的細(xì)胞區(qū)分開(kāi)來(lái)。在雙變量流式細(xì)胞儀的散點(diǎn)圖上,若橫坐標(biāo)表示Annexin V信號(hào),縱坐標(biāo)表示PI信號(hào),則左下象限 (Annexin V-/PI-) 代表活細(xì)胞;右下象限(Annexin V+ /PI-) 代表早期凋亡細(xì)胞;右上象限(Annexin V+ /PI+)代表凋亡晚期細(xì)胞和壞死細(xì)胞;左上象限(Annexin V-/PI+) 被認(rèn)為是許可范圍內(nèi)的檢測(cè)誤差。
